The bianconeri got off to the perfect start with Marco di Vaio firing them in front after just eight minutes.
The former Parma man netted his first league goal for Juve with a simple finish from Alessandro Del Piero's pass.
The home side doubled their lead in the 20th minute with a rare goal from French defender Lilian Thuram after a brilliant counter attack.
Juve broke from their own box defending a Milan corner to devastating effect with Del Piero again being the architect of the goal setting up Thuram for the powerful shot, which beat Christian Abbiati.
Milan fought their way back into the game and gave themselves a lifeline with Andrea Pirlo pulling a goal back from the penalty spot just past the half hour mark.
Juve defender Alessandro Birindelli was adjudged to have handled the ball in the area and a penalty was awarded from which Pirlo confidently scored.
The second half was less open with Juve content to defend their lead and they held out against a spirited Milan fightback.
The win lifts Juve into second spot just a point behind leaders Internazionale while Milan fall to third spot.
