Leno, 21, has become a regular starter at the BayArena since his move from Stuttgart in the summer of 2011.
His previous deal ran until June 2017, but head coach Sami Hyypia and the club's sporting director Rudi Voller were determined to hang onto the promising youngster.
"Bayer Leverkusen turned me from a 19-year-old third-division goalkeeper into a number one at a Champions League club," Leno said.
"That act of faith made a great impression on me and I'm incredibly thankful for the club for that."
Meanwhile, Voller said: "We have been working hard on Bernd's contract extension for a long time. The lad is one of the best keepers in Germany as he proves week after week."
Leverkusen currently sit third in the Bundesliga - four points behind leaders Bayern Munich - and are also in contention to qualify for the knockout stages of the Champions League.
They host Group A leaders Manchester United in the next round of fixtures and victory would put them on the brink of reaching the last 16.
