Poulsen is struggling to be ready for the clash against champions Werder Bremen on Friday August 7 after jarring his knee in training.
The Denmark international is set to miss the Intertoto Cup meeting with Esbjerg on Wednesday and is concerned he will be sidelined for a couple of weeks.
"I twisted my left knee during the second training session after the vacation," Poulsen told BT.
"My knee has been X-rayed but nothing was hurt.
"There is some fluid in the knee, therefore I don't expect to start practicing until during the course of the week.
"Besides the two clashes against Esbjerg, we have just got one more friendly and I haven't started training yet."
Meanwhile, Esbjerg coach Ove Pedersen is relishing the prospect of tackling Schalke in the Intertoto Cup.
Esbjerg reached the semi-finals of the competition with a 5-1 aggregate victory over Lithuanian outfit FK Vetra and Pedersen can see a spot in the Uefa Cup on the horizon.
Pedersen told Ritzau: "I would rather like to focus on the fact that we are in the Intertoto Cup semi-final and are close to a place in the Uefa Cup."
