The promoted side threatened to cause a shock when Mathias Abel and Conor Casey had them in command after 17 minutes.
But Petrov scored twice in first half injury time before Mainz were reduced to ten men when substitute Jurgen Krammy was sent off for deliberate handball on 57 minutes.
Petrov slotted home the resulting penalty and notched again on 62 minutes. Christof Babatz pulled one back on 75 minutes but Wolfsburg clung on for the spoils.
Bayern Munich suffered a surprise 2-0 defeat at Borussia Monchengladbach, which enabled the home side to move out of the relegation zone.
Marcelo Pletsch broke the deadlock on 49 minutes and Joris van Hout sealed the success on 83 minutes, just five minutes after coming on as a substitute.
Reigning champions Werder Bremen lost ground on the leaders as they had to come from behind to earn a point at home to Hamburg.
David Jarolim had opened the scoring for the visitors on 22 minutes but Christian Schulz snatched a point with 15 minutes remaining.

Schalke withstood a strong fightback to hang on for a 3-2 triumph at home to Stuttgart with all the goals coming inside the opening 33 minutes.
Ailton and Levan Kobiashvili gave Schalke a dream start by scoring in the opening two minutes and Lincoln made it 3-0 on 25 minutes.
Zvonimir Soldo and Imre Szabics scored within three minutes of each other to hand Stuttgart hope but they could not force an equaliser in the second period.
Hertha Berlin produced a solid performance away from home to beat Freiburg 3-1, despite the visitors finishing with ten men.
Ellery Cairo put the hosts ahead on 25 minutes but Hertha were level before the interval courtesy of Nando Rafael.
Gilberto and Arne Friedrich scored after the break for Hertha, who were reduced to ten men when Alexander Madlung was dismissed on 79 minutes.
Hannover cruised to a 3-0 home win over Bochum with Thomas Christiansen netting first after 16 minutes.
Second half efforts from Daniel Stendel and Leandro wrapped up the points for Hannover.
