The Royal-Blues had to settle for a 1-1 first leg draw against the Ukrainian outfit as they conceded a late goal to Brazilian striker Brandao.
The German side opened the scoring after just seven minutes when Ailton fired home with his left foot after a clever chip from Ebbe Sand, and Rost denied the home side until the 86th minute.
"They fully deserved the equaliser as disorder ruled in our defence," fumed Rost.
"I am angry about conceding a goal after 86 minutes, but we never controlled their fast strikers and could have been set for a big defeat."
Schalke coach Ralf Rangnick, who was without playmaker Lincoln and influential defender Marcelo Bordon, was less critical of the side, but hailed his keeper's performance.
"Frank showed some excellent reflexes and denied the equaliser for most of the match, he has been on top form for several months now," said Rangnick.
"The result was fair, although we also had some chances to double our lead towards the end.
"A 0-0 would do for us in the rematch and this means that Shakhtar will be forced to take some action and this should give us the chance to put pressure on their defence."
