PSV, last season's runners-up, are embarking on their next chapter under new coach Philip Cocu, and things looked to be going well at the Kyocera Stadium as they raced to a 3-0 lead in 33 minutes.
Georgino Wijnaldum got the ball rolling after 12 minutes before a Danny Holla own goal (27) and Jetro Willems' 34th-minute strike gave the scoreline a one-sided look.
Holla then made amends for his earlier blunder by pulling one back for ADO at the right end on the stroke of half-time and a late effort from Mike van Duinen set up a nervy finish, but PSV hung on.
FC Groningen looked more confident as they hammered NEC 4-1 in Nijmegen.
Tjaronn Chery (10) and Andraz Kirm (27) put the visitors in the driving seat with Navarone Foor replying for NEC before the interval.
Second-half strikes from David Texeira (54) and Richairo Zivkovic (90) put the result beyond doubt, however.
There were also goals aplenty at Abe Lenstra Stadion where 10-man Heerenveen beat AZ Alkmaar 4-2.
Alfred Finnbogason's brace (22 and 54) sandwiched Aron Johannsson's effort (50) as the home side took a 2-1 lead, which they extended through Pele van Anholt after 56 minutes.
The 72nd-minute dismissal of Mitchell Dijks did not help Heerenveen's cause and they conceded another Johannsson goal two minutes from time, but the players responded and Rajiv Van La Parra wrapped up the points in stoppage time.
FC Twente, who fell off the pace towards the end of last season to finish sixth, opened with a goalless draw at home to RKC Waalwijk.
