The Bayern Munich man limped out of training with thigh trouble on Monday and will miss the clash at the Westfalenstadion in Dortmund along with club team-mate Jeremies.
The midfielder had not played for Germany for nearly 17 months while he recovered from a lengthy knee injury before the former Hertha Berlin ace made his international comeback in last weekend's 0-0 draw against Iceland.
Carsten Ramelow missed Monday's training session and is also a doubt for the clash against the Scots with a thigh problem and Deisler's injury leaves Voller short of creativity in midfield with Michael Ballack expected to now burden that role by himself.
If Ramelow is also forced out of the encounter, Voller may have to go into the crunch game with inexperienced duo Andreas Hinkel and Michael Hartmann in a game both need to win to close the gap on Group Five leaders Iceland.
Voller has already been forced to rebuild his successful World Cup formation with long-term absentees such as Liverpool ace Dietmar Hamann, Torsten Frings,Christoph Metzelder and Jens Nowotny, but the latest developments leave him dramatically short of options in midfield.
