The Hearts stopper has been striving to shake off a virus this week to ensure he would be fit for Berti Vogts' side.
But the powerful centre half has been forced to concede defeat in his quest to regain his fitness ahead of Saturday's clash against Norway.
Pressley's absence merely compounds Vogts' problems in defence, with Gary Caldwell admitting his compatriot will be missed.
"It is difficult. Elvis (Pressley) and Andy Webster had a good partnership in defence and they obviously know each other well," said Caldwell.
"But we have today and tomorrow to work on it and get it right. We will be ready for Saturday's game."
Vogts is already without the likes of Christian Dailly and Malky Mackay for the games against Norway and Moldova which could be decisive in Scotland's attempts to reach the tournament in Germany.
The Scotland coach had already drafted in Stephen Caldwell and Graeme Murty as cover but must now decide if he needs additional back-up in Pressley's absence.
