Tacchinardi limped out of the side's final training session on Friday with a thigh problem and he is a major doubt for the game in Milan.
Tests have shown no major injury to Tacchinardi, but Trapattoni will not risk him unless he is 100 percent fit and he is keeping his options open as who to play should the Juve star miss out.
"I won't risk Tacchinardi if he has a fitness problem," said Trapattoni.
"Cristiano Zanetti and Perrotta are now favourites to play in central midfield, with Gennaro Gattuso as a very valid alternative to be considered."
Meanwhile, Christian Vieri has declared himself fit to face Wales after recovering from a knee knock.
The Internazionale striker has missed some training this week, but he is feeling fit and is ready to play.
"I feel good," said Vieri. "That apprehension has passed by and thankfully so has the pain."
"We have performed tests and the tendon is clear.
"This is reassuring for me and the team. Everything is just fine and I will start against Wales."
