The 24-year-old, who won the 2008 US Women's Open, carded her second successive five-under 67 at Mission Hills in California to pull clear of the field.
Park started the day just one shot ahead of American Lizette Salas and the pair both birdied the opening hole before Salas took a share of the lead with another birdie at the second.
A long birdie putt at the fifth put Park back in front and she remained in charge after Salas fell back with a bogey on the eighth.
Further birdies at the 10th, 12th and 17th helped extend Park's advantage over Salas, who finished with a 69 to retain second place.
It appears to be a two-horse race for the title as it is then three shots back to a group of six players on six under: Norway's Suzann Pettersen (67), Americans Angela Stanford (66) and Jessica Korda (68), Australian Karrie Webb (67), France's Karine Icher (68) and Thailand's Pornanong Phatlum (70).
British hopes of success were dashed when first-round co-leader Jodi Ewart Shadoff, who had remained in contention after a level-par second round, fell away to a share of 17th place on two under after a two-over 74.
The Englishwoman is one shot ahead of Scotland's Catriona Matthew who had her best round of the week, a two-under 70.
Park said: "I feel really about my swing and my stroke at the moment and am really feeling comfortable around this golf course.
"I've had a few good rounds and I just need one more tomorrow.
"Winning this one would mean a lot. It's a tournament I have always wanted to win with the special ceremony and jumping in the water.
"Everybody just wants to do that I think and it feels really special."
