The World No. 13 had an eagle and five birdies in a seven-under par round of 64 at Grey Silo, which took her to 15-under and gave her a three-stroke lead over America's Angela Stanford.
Stanford began the day tied for the lead with Matthew and had five birdies and a bogey in a 67 to reach 12-under.
Sweden's Anna Nordqvist (64) is tied with four other players for fourth, at 11-under, while World No.1 Inbee Park, fresh from winning her third major title of the year a fortnight ago, celebrated her 25th birthday with a round of 67 to finish tied for eighth on 10-under.
Good round from Pettersen
Norway's Suzann Pettersen is also in that group after a fine round of 64, which featured eight birdies.
Matthew got things going with an eagle at five, where she converted a 25-foot putt, and she picked up further shots at six, eight, 11 and 16, before finishing in style by sinking a four-foot birdie putt at the last.
"I played really well again and gave myself a lot of chances," she said. "It feels really good. I've holed a lot of putts. I've given myself a lot of chances, so I wouldn't say I've really holed many super long ones but, every time I give myself a chance, I'm kind of taking advantage of it.
"When you're holing putts, it all seems quite easy."
Park is chasing a fourth LPGA win in as many starts and had 'happy birthday' sung to her by spectators at the 18th.
"I've never had that many people wishing me happy birthday," she said. "I'll remember my 25th birthday forever."
