The Australian bogeyed the 13th and 15th holes but cancelled that out with two birdies after the turn which, added to a net gain of three shots on the front nine, left her with a three-under par effort of 69 on the day.
She sits on nine-under for the tournament, two shots clear of 2012 champion Angela Stanford, who also carded a second round of 69 at Sentosa Golf Club in Singapore.
Sweden's Caroline Hedwall had looked like she could challenge Webb at the top of the leaderboard but two bogeys and a double bogey on her final three holes left with only a share of fourth place.
After the round, the 39-year-old Webb was keen to point out that her wealth of experience is proving to be a help rather than a hindrance against some of the game's rising stars.
"Did you ask any of the younger players if it's hard to play against girls in their 30s? Because you always ask me the opposite question," the seven-time major winner said.
But she added: "All these young players coming up are athletes. For me, I've had to learn to get in the gym and do the work required."
"My workouts have just gradually increased so it wasn't hard-core to start with, where I would have just hated it and never done it."
Taiwan's Teresa Lu is three strokes behind Webb in third place, while American trio Paula Creamer, Morgan Pressel and Danielle King joined Swedes Hedwall and Anna Nordqvist in fourth place on four-under.
