The 23-year-old, who is making only her third LPGA Tour start - and first in the United States, started the day one shot clear of American Morgan Pressel after opening with a 64 at Phoenix's Wildfire Golf Club.
Lee began with a birdie but slipped up with her lone bogey at the eighth before birdieing the 11th and 13th and then eagling the par-five 15th to double her advantage on 13-under 131 after 36 holes.
New Zealand teenager Lydia Ko, 16, is Lee's nearest challenger on 11 under after carding a 66 which also included an eagle at the 15th.
Korean Yoo Sun-Young is third on 134 after a 65 with American Jessica Korda and South Koreans Chella Choi, Ryu So-Yeon and Park In-Bee - the world No 1 - sharing fourth on 135.
England's Jodi Ewart Shadoff fired a 69 for a share of eighth place on 136, while Pressel is a shot further back after struggling to a 72.
Lee said: "I played a little boring on the front nine so I went more aggressive on the back nine.
"It worked well so now I plan on being aggressive the rest of the week."
Ko, who like Lee is seeking her first LPGA title as a professional said: "It's great to see two rookies out there. Mirim's obviously playing really good golf and there are a lot of birdies out there.
"I've never really met her before so should be good to get to know another rookie."
