Having managed a 63 on Friday the Argentine went one lower 24 hours later, nine birdies on his card moving him to 20-under for the tournament.
Oliver Fisher sits alone in second place after signing for a 66, the Englishman covering his final four holes in four-under to quickly erase the memory of three earlier bogeys.
England's Richard Bland (64), and Thomas Aiken (66) of South Africa are a further two shots adrift alongside overnight leader John Hahn, the American slipping back after an even-par 71.
A strong wind made scoring tougher for many on Saturday, though it didn't stop Grillo who eagled the 72nd hole to finish second in the recent Dubai Desert Classic, from producing something special.
He picked up five shots in the space of six holes to reach the turn in 31 shots, then took the same number on his way back in thanks to birdies at the 12th, 13th, 15th and 16th.
By the time he came off the course it seemed he would take a healthy cushion into the final round, only for Fisher to close the gap late on.
Having birdied the 15th, the 25-year-old Londoner carded an eagle at the short par-four 16th, his drive bouncing through a greenside bunker. He then holed from off the green on the last, meaning for the second successive round he'd taken 31 shots on the back nine.
