The South Korean recovered from a wobbly start, recording his first bogey of the week at the opening hole, to shoot a score of 71 and finish two clear of American duo Andrew Svoboda and Robert Streb.
Noh had started the day with the same cushion over Keegan Bradley, only to immediately get into trouble off the tee to suffer an early blemish on his card.
However the 22-year-old showed maturity beyond his tender years to recover from the setback. He gained the dropped shot back at the eighth, then added another birdie at the 10th.
Although he did have bogeys at the 12th and 15th, on each occasion he responded with a birdie at the next.
With no-one able to put him under real pressure Noh was able to tee off at the final hole with a two-shot cushion. He played it safe by aiming away from the water, allowing him to make a comfortable par five to end on 19 under.
His triumph means Noh becomes the fourth South Korean to triumph on the PGA Tour - he also secures places in the field at this year's US PGA Championship and the 2015 Masters.
Jeff Overton finished alone on 16 under, two clear of fellow Americans Robert Garrigus - who fired a stunning round of 64 that saw him manage four birdies and an eagle after the turn - Erik Compton and Charley Hoffman.
Englishman Justin Rose ended on 13 under, the same score as Bradley after the American saw his hopes of challenging disappear with a disappointing 75.
The former US PGA Champion was level for his round after five holes but a triple-bogey on the sixth effectively ruined his chances of victory. After putting his tee-shot in the water, Bradley was on the green in four but then three-putted to tumble down the leaderboard.
Paul Casey managed a 73 to finish on 12 under while Padraig Harrington finished one under for the tournament at TPC Louisiana.
