Vinales, the reigning Moto3 world champion, carved his way through the field after starting in sixth position on the grid to take victory by 4.009sec from pole sitter Esteve Rabat.
Kalex rider Vinales finished fourth in the season-opener in Qatar - a race which was won by his compatriot and fellow Kalex rider Rabat.
Suter rider Dominique Aegerter of Switzerland finished third ahead of Mika Kallio in fourth and Simone Corsi in fifth.
Britain's Sam Lowes could only manage 16th place after getting caught up in a crash at the first corner while fellow Brit Gino Rea finished down in 25th.
The Moto3 race saw a second successive season win for 19-year-old Australian Jack Miller.
The KTM rider, whose maiden race win came in Qatar last month, had to fend off a challenge from Romano Fenati, who was second, and Efren Vazquez who finished third.
"I felt strong on the brakes," said the Queenslander. "That's always been one of my strong points, maybe too strong as it can kill the exit speed.
"But here it works really well. I'm stoked to have won, I led every lap."
Britain's Danny Kent finished eighth on the Husqvarna, one place ahead of Honda rider and fellow Brit John McPhee.
