The veteran Australian will lead out Brian McClennan's side on his home ground at Warrington's Halliwell Jones Stadium on Friday, 14 June.
The 35-year-old full-back led the Exiles to a 32-20 victory in the fixture at Huddersfield last July and was the first name included in McClennan's 27-man squad.
McClennan said: "Brett Hodgson brings a lot to the side on and off the field. His leadership and experience is going to be crucial for us. It's a one-off match and we're going to give it our best shot."
Hodgson, who will be joined by Warrington team-mates Joel and Michael Monaghan and Trent Waterhouse, added: "The Exiles is a wonderful concept.
"We have an exciting squad and it's a great honour to be chosen as captain - especially in front of my home supporters. There is no second game this year, so everything is riding on the one match.
"For players on either side it's their only representative opportunity before the Rugby League World Cup, so I expect a very intense battle.
"I really hope the Warrington public come out and support the match in huge numbers. It's important that England are at their best at the World Cup and they are going to have to be that to beat us."
St Helens also have four players in the squad, including Exiles regulars Willie Manu and Sia Soliola, but winger Francis Meli has pulled out with a dislocated elbow.
Among the newcomers are Hull KR second rower Cory Paterson and Wigan's former Rovers stand-off Blake Green, who looks set to renew his old half-back combination with Michael Dobson.
Exiles' squad: Louis Anderson, Daryl Millard, Zeb Taia, Chris Bailey, Antonio Kaufusi, Michael Dobson, Mickey Paea, Cory Paterson, David Faiumu, Blake Green, Pat Richards, Rhys Hanbury, Brett Hodgson (capt), Joel Monaghan, Michael Monaghan, Trent Waterhouse, Lance Hohaia, Willie Manu, Tony Puletua, Iosia Soliola, Kylie Leuluai, Joel Moon, Heath L'Estrange, Manase Manuokafoa, Jarrod Sammut, Mark O'Meley, Justin Poore.
