The 33-year-old Australian prop, who played for the Giants between 2011 and February 2013, spent most of this year with National Rugby League side Sydney Roosters.
Despite being suspended by the club for breaching their code of conduct shortly after arriving, he played 20 times as the Roosters won the NRL title.
The 26-18 Grand Final victory over Manly Sea Eagles is the final game of his 215-match career.
"I've had some time to reflect and think about my future since the Grand Final and this definitely hasn't been an easy decision, but it's the right one," said O'Donnell.
"I'm so grateful for everything rugby league has given me. Each of the clubs I've played at have made me a better man and I've made so many friends along the way."
O'Donnell, who also played club rugby for Wests Tigers and North Queensland, played five times for New South Wales and won 11 Test caps with Australia.
