Leeds landed a home tie after an impressive 60-6 fourth-round win at neighbours Wakefield on Sunday.
But Saints were forced to dig deep for their place in last 16, snatching a 17-16 win over Huddersfield Giants with Luke Walsh's last minute drop goal.
Nathan Brown's men face another testing trip to Yorkshire as they pursue their first Wembley triumph since 2008.
"At the end of the day you've got to beat them to get there," said Brown.
"We've played a top side today and going to Leeds will be another great challenge for us."
Holders Wigan face part-time opposition for the second time as their 58-6 win over Dewsbury has set up a home clash with Championship One team Hunslet.
Salford, who are expected to unveil Iestyn Harris as their new head coach on Monday, have been drawn against Widnes, while Bradford will host Catalan Dragons in the other all-Super League tie.
There will be at least two Championship clubs in the quarter-finals, with Leigh meeting Featherstone and Swinton playing Keighley in the fifth round.
Fifth round draw:
Wigan v Hunslet
Leigh v Featherstone
Leeds v St Helens
Warrington v Doncaster
Salford v Widnes
Bradford v Catalan Dragons
Swinton v Keighley
Castleford v Sheffield Eagles
Ties will be played on the weekend of April 26-27th.
