The Rhinos won last year's semi-final against the Warriors 13-12 before seeing off the Warrington Wolves and they now have to do it again 12 months later, despite finishing above Wigan in the table.
McGuire was Leeds' hero last week after he kicked the decisive drop-goal that gave Brian McDermott's men a slender 11-10 triumph over bogey side St Helens at Headingley.
Leeds edged out Wigan 20-6 in Super League earlier this month, however, McGuire knows Shaun Wane's men will be a different prospect on Friday with the incentive of a trip to Old Trafford on the horizon.
"It's going to be very tough. Wigan are quality team. They have been very consistent this season. They have some great players," McGuire told Sky Sports.
"I think we played them three or four weeks ago and we had a very tough tussle with them and only just came out the other side with a win.
"We're going to be very confident and we're only one game away from a Grand Final which makes the game even more exciting. Everyone from our camp is looking forward to it."
Leeds' vastly experienced prop Jamie Peacock has been nominated for the 2013 Man of Steel award and McGuire believes his team-mate would be a worthy winner having claimed the prize for the Bradford Bulls ten years ago.
"He's been massive for us this season," McGuire continued. "We've had a lot of injuries, but JP has been there throughout and carried the team at times on his shoulders.
"He's been unbelievable and he'd be very worthy of the Man of the Steel tag. He's been a great leader for a number of seasons now and I think personally he would be my choice of Man of Steel."
