The 32-year-old centre or stand-off joined up with the Tomahawks on their return from Toulouse, where they stunned France by beating them 22-18 in a warm-up match on Friday.
But the team has now announced his withdrawal ahead of their opening game against the Cook Islands on 30 October.
"Ryan McGoldrick has been granted leave from the USA Tomahawks' Rugby League World Cup camp due to personal reasons," read an official statement.
"McGoldrick is a valued member of the Tomahawks and has worked tirelessly to promote this tournament and USA rugby league.
"We look forward to seeing him in the red, white and blue again in the future."
The loss of the experienced McGoldrick is the second blow to hit the USA camp after it was discovered second rower Mark Cantoni broke an arm in the victory over France.
"Mark's been working for two years to get this opportunity so it's disappointing for him," said Tomahawks coach Terry Matterson.
"Hopefully we may be able to get him a run at the back end, that's how keen he is."
The Tomahawks have decided not to call up a replacement, although former Hull KR forward Clint Newton and Wigan's new signing Eddy Pettybourne come into the squad after missing the match in Toulouse.
