Having been cursed by injury in 2012, the 22-year-old will end the year on the sidelines. It was decided best not to risk the persistent issue with his hamstring on the four-match end-of-season tour.
O'Connor on his Twitter page said: "[Just] had meeting with doc, looks like I'll be in #Teamrehab til after spring tour. Gutted I can't play but no risks with huge year ahead #2013."
The players' absence was also confirmed on the official Twitter page of the Australia Rugby Union, highlighting even more injury troubles for the team.
The playmaker has not played a Test this year due to liver and hamstring problems, which also ruled him out of the majority of the Super 15 season for the Melbourne Rebels.
Ahead of the tour, which begins on November 10 against France in Paris, Australia have further injury doubts with skipper James Horwill (hamstring), Berrick Barnes (lung) and David Pocock (knee) all in some doubt of missing the trip.
