The Richmond centre is part of a youthful back-line, as several of the regulars are away in Houston for the Sevens World Series.
Ceri Large and Georgina Gulliver start as the half-back pairing, while Chamberlain is joined in the centres by Amber Reed. On the wing Lydia Thompson is paired with Kay Wilson, who plays internationally for the first time in nine months, while Lauren Cattel completes the back three.
England have opted for a much more experienced pack with captain Sarah Hunter starting at No 8. She is joined in the back row by Hannah Gallagher and Jo McGilchrist, who starts her first game in 17 months.
In the front-row, hooker Emma Croker is joined by Roz Crowley and the inexperienced Kate Newton, who will be starting just her third game for England. Tamara Taylor and Emily Braund complete the line up in the second-row.
Long-term absentees Fiona Pocock and Rebecca Essex return to the bench. Harriet Millar-Mills is also named among the replacements and could face her sister, who is due to line up for the Scots.
England head coach Gary Street said: "This year's Six Nations is an exciting challenge for me and Graham Smith. Training has gone well with several new faces in the squad and we are looking fresh and the team are really up for this.
"Many of our players may only have a handful of caps between them but the likes of Amber Reed, Hannah Gallagher and Ceri Large have all played France and the world champions New Zealand, and have beaten them.
"This is a big year for Scotland with RWC qualification at stake so we are not going to be taking anything for granted. They have got some very strong forwards and an excellent captain in Susie Brown so Saturday's opener is definitely going to be a tough test."
