Bath hooker Rob Webber has been given the honour of leading the side with 21-year-old Matt Kvesic and Marland Yarde set to pull on an England jersey for the first time.
Gloucester-bound forward Kvesic and London Irish winger Yarde are two of nine players under the age of 24 in the England starting XV.
Wasps winger Christian Wade was voted players' player of the year earlier in the week and features on the right wing as the only other uncapped starter.
Other newcomers named among the replacements for the non-cap game are Bath centre and former England rugby league international Kyle Eastmond, former Sale lock Kearnan Myall, Exeter scrum-half Haydn Thomas, Sale prop Henry Thomas and Wasps No 8 Billy Vunipola.
"We've selected a young and exciting side and this is a great opportunity for these players who have shone in the Aviva Premiership this season to step up in front of passionate home crowd at Twickenham Stadium," England Head Coach Stuart Lancaster said.
"Taking on a very experienced Barbarians team is the first challenge they face this summer with matches in Uruguay and Argentina to follow and this sequence of games is a key part of our development as a team.
"Rob Webber finished the season strongly with Bath and I know his leadership qualities well. I am sure he'll show the way backed up by the other senior players in the team."
Meanwhile, former England international centre Mike Tindall will face his compatriots at Twickenham as he leads the Barbarians XV.
Tindall has been partnered with Casey Laulala in midfield while Welsh pairing Dwayne Peel and James Hook will start at half-back.
Young Wasps back Elliot Daly will start at full-back with teammates Jonathan Poff and Marco Wentzel in the forwards and Sam Jones completing the capital quartet having been named on the bench.
Up front the pack includes French international Imanol Harinordoquy, Alessandro Zanni, Jim Hamilton, James Johnston and Andrea Lo Cicero.
