The Azzurri fell to their second successive loss in this season's tournament on Saturday with a 26-9 defeat at the hands of Wales at Rome's Stadio Olimpico.
The hosts, who upset France in a memorable opening match, were undone by second-half tries from Jonathan Davies and Alex Cuthbert.
And Italian pain was not confined solely to the result, with half-backs Kristopher Burton and Edoardo Gori both suffering injuries in the rain-sodden contest.
Fly-half Burton has a problem with his right ankle, while Gori stretched medial ligaments in his knee.
The news has given Brunel cause for concern as he also continues to be without suspended skipper Sergio Parisse.
Parrisse, who is serving a 30-day ban after being sent off for insulting the referee during a French club game for Stade Francais, is likely to appeal the punishment, but as things stand he is likely to be suspended until March 18.
Martin Castrogiovanni stepped in to replace the talismanic captain for the defeat against Wales while veteran back-row forward Mauro Bergamasco was called into the squad as cover.
