The world champion was thrown over his bike after a collison with Panthers guest Mads Korneliussen in an early heat of the KO Cup semi-final second leg.
Hospital scans revealed the Australian's injuries were worse than originally feared, having sustained a broken tibia, broken rib and knee ligament damage.
The Poole Pirates ace will undergo further scans in hospital, and he is expected to require extensive surgery over the next week.
Korneliussen, who unwittingly caused the crash when he slid into Holder's path, was withdrawn from the fixture but escaped serious injury.
Despite losing Holder, the Pirates managed to scrape into the final after Krzysztof Kasprzak and guest Niels-Kristian Iversen took a 5-1 in a last-heat decider to win by five points on aggregate.
