Chris Coyne fired Luton in front on 29 minutes and Steve Howard doubled the lead after 54 minutes.
Kevin Nicholls wrapped up a comfortable victory for Mike Newell's men with a penalty on 74 minutes.
Hull drop to second after being held to a surprise draw at home to Peterborough.
Steve Thomson gave Peterborough a shock lead on 26 minutes, but Hull turned things around with goals from Nick Barmby (46) and Stuart Green (60) 15 minutes after the restart.
Callum Willock popped up in the final minute to deny Hull all three points and earn a crucial draw for Peterborugh.
Tranmere kept pace with the top two with a thumping 5-1 win at troubled Wrexham.

Eddie Hutchinson grabbed the all-important winning goal for the visitors on 29 minutes.
Peter Clarke's late goal earned Blackpool a 2-2 draw at Swindon.
Keigan Parker gave Blackpool the lead after 18 minutes, but Swindon hit back with two goals from Sam Parkin before half time (36, 45).
That looked like being enough for Swindon until Clarke pounced in the dying seconds.
Milton Keynes Dons climbed out of the bottom four with a 2-0 win over Colchester.
Ben Harding gave MK Dons a dream start with the opener after just two minutes and Wade Small made sure of victory with the second on 18 minutes.
Doncaster brought FA Cup heroes Oldham back down to earth with a 2-1 win at Boundary Park.
Gregg Blundell got Doncaster off to a flying start with a goal in the very first minute and Jermaine McSporran added a second nine minutes from time.
Scott Vernon pulled a goal back six minutes later but it was nothing more than a consolation for the hosts.
Barnsley survived a scare before coming out 4-2 winners over Huddersfield in the Yorkshire derby at Oakwell thanks to a hat-trick from Michael Chopra.
Barnsley raced into a three goal lead with strikes from Michael Chopra (9),(24) and Michael Boulding (38).
Tykes goalkeeper Ross Turnbull was sent off on 54 minutes and Pawel Abbot quickly reduced the arrears from the penalty spot.
Andy Booth pulled back another on 70 minutes only for Chopra to make sure of the win with his hat trick three minutes from time with a penalty.
Walsall recorded their first win in five games with a 3-0 success over Chesterfield.
Darren Wrack gave Walsall the lead on 14 minutes and Jorge Leitao doubled the advantage six minutes later.
