Mathieu brushed aside the Italian qualifier 6-3 6-4 to move into the last eight of an event which has been severely affected by the withdrawal of a host of big names.
Defending champion Philipp Kohlschreiber was forced to miss the clay-court event while former world number two Tommy Haas withdrew on Tuesday because of a recurrence of a shoulder injury.
World number 19 Mathieu will face eighth seed Marin Cilic in the quarter-finals - the Croat beat Germany's Philipp Petzschner earlier in the day.
Wildcard Marat Safin moved into the second round with a 6-3 3-6 6-4 victory over Carlos Berlocq.
Safin, the former world number one who has tumbled down the rankings after a series of injury problems, faces Michael Berrer of Germany next.
The Russian is on course to meet Chilean second seed Fernando Gonzalez in the last eight.
