The New England Patriots ended the Baltimore Ravens unbeaten start to the season with a 27-21 victory.
Quarterback Tom Brady passed for one touchdown and ran in another himself as New England improved their own record to 3-1 - now the same as their opponents.
Meanwhile, brothers Peyton and Eli Manning ensured the Indianapolis Colts and the New York Giants retained their 100 percent records, the quarterback siblings leading their sides to victories over the Seattle Seahawks and the Kansas City Chiefs respectively.
The Cincinnati Bengals ensured Cleveland remain winless as Shayne Graham kicked a last-gasp field-goal to sink the Browns in overtime.
Elsewhere in week four of the regular season, there were also victories for the Houston Texans, the Jacksonville Jaguars and the Washington Redskins.
Tom Brady and New England ensured Baltimore's perfect start came to an end with a hard-fought 27-21 victory over the Ravens at Gillette Stadium.
Brady edged his side 10-7 ahead on a one-yard run in the second quarter and then found Randy Moss with a 14-yard scoring pass in the third to open up a 24-14 lead.
However, Baltimore refused to lie down and had the chance to force the game into overtime, but wide receiver Michael Clayton let a fourth-down pass from Joe Flacco bounce off his chest and fall incomplete with just 28 seconds left on the clock.
Peyton Manning threw for over 300 yards for the fourth game in succession as the Indianapolis Colts thrashed the Seattle Seahawks 34-17 to move to 4-0 for the season.
It is the first time in his illustrious career that Manning has achieved such a run and the Colts have now won 13 consecutive regular-season games - the longest run in the league.
Without the services of first-choice quarterback Matt Hasselbeck the Seahawks were never really in the game and managed to add a touch of respectability to the scoreline with a couple of fourth-quarter touchdowns.
Manning threw for a total of 353 yards and two touchdowns, while Joseph Addai and Donald Brown ran in a touchdown apiece.
Eli Manning threw three touchdown passes before giving the Giants an injury scare by leaving in the fourth quarter of their 27-16 win over the Kansas City Chiefs.
The quarterback hit receiver Steve Smith, who finished with 134 yards on 11 catches, twice in the end zone before teaming up with Hakeem Nicks on a 54-yarder in the third quarter.
That score put the Giants 27-3 up and, with Manning taken out of the game, the Chiefs - now winless in eight - gave their fans something to cheer about with two late touchdowns, both short passes thrown by Matt Cassel.
Their only points before then had been a Ryan Succop field goal from 34 yards out after Tamba Hali had forced a turnover on a sack of Manning, who left the game with what was described as a bruised heel.
The Lions returned to losing ways in Chicago as they were blitzed by the Bears, who have now won three straight, at Soldier Field.
Despite only throwing for 141 yards, quarterback Jay Cutler managed touchdown passes to tight ends Kellen Davis and Greg Olsen after running into the end zone himself in the first quarter.
Matt Forte also galloped in for a score, one of three the Bears managed on the ground, as the running back returned to form with 121 yards on 12 carries.
Chicago's special teams also came up with a TD, receiver Jonny Knox taking a kick-off back 102 yards. Despite a pair of Kevin Smith scores, Detroit (1-3) were unable to follow-on from their win over Washington.
Cincinnati came out on top in the battle of Ohio against Cleveland thanks to a Shayne Graham field goal with four seconds left in overtime.
The Bengals had led 14-0 at one stage thanks to the first of Chad Ochocinco's two touchdowns and a 75-yard fumble return by Robert Geathers after Jerome Harrison, starting in place of the injured Jamal Lewis, was stripped of the ball.
Cleveland came roaring back, though, quarterback Derek Anderson - now starting in front of Brady Quinn - throwing for one TD and also running in for one himself from a yard out. A pair of Billy Cundiff field goals then put the Browns 20-14 up.
Carson Palmer teamed up with Ochocinco again to level the scores but with the point after blocked, extra time was required. Just as a tie looked likely Graham held his nerve to knock over a 31-yarder to win it.
Steve Slaton notched two touchdowns as the Houston Texans eased to a 29-6 victory over the struggling Oakland Raiders in Texas.
The running back ran in from 32 yards out in the second quarter and then reeled in a scoring pass from Matt Schaub soon after to help Houston lead 20-6 at half time.
The hosts added a saftey when Oakland tail-back Justin Fargas was tackled by Brian Cushing in the end zone before Jacoby Jones rounded out the scoring with 95-yard kick-off return.
All Oakland, who rushed for 45 yards, managed in reply was a pair of Sebastian Janikowski field goals.
Jacksonville piled on the misery for Tennessee as David Garrard enjoyed an excellent day under centre in a 37-17 mauling of the winless Titans.
Quarterback Garrard twice found Michael Sims-Walker in the end zone in the second quarter before throwing a 33-yarder to tight end Marcedes Lewis late on. He finished 27-for-37 for 323 yards.
Maurice Jones-Drew also scored on the ground for the Jags, who managed 442 yards on offense. Josh Scobee knocked over field goal attempts from 22, 33 and 50 yards to help pad out the final score.
Kerry Collins threw for one touchdown and ran for another but by then the Titans, without cornerbacks Cortland Finnigan and Vincent Fuller through injury, were already on their way to a fourth successive loss.
Jason Campbell engineered a third-quarter comeback to help the Washington Redskins avoid another upset, this time at the hands of Tampa Bay.
The quarterback was intercepted twice, as well as losing a fumble, in the first half but bounced back in style, throwing TD's to tight end Chris Cooley and wide receiver Santana Moss.
The Buccaneers had led 10-0 at half time, a Mike Nugent field goal following on from Antonio Bryant's eight-yard pass from new quarterback Josh Johnson.
However the Redskins - beaten by the Lions last time out - rallied with 16 points in under nine minutes, Campbell's two touchdown throws being added to by a 42-yard field goal from Shaun Suisham.
Although Nugent did kick a 22-yarder to close the gap to three points that was as close as Tampa managed to get, meaning they are now 0-4 for the season.
The New York Jets' unbeaten start ended in New Orleans as the unbeaten Saints pounced on rookie quarterback Mark Sanchez in a 24-10 victory at the Superdome.
Sanchez saw an attempted touchdown pass intercepted close to the Saints goal line at the start of the second quarter - Darren Sharper then running the length of the field to complete a 99-yard touchdown.
He then surrendered the ball in his own endzone just before half time, with Remi Ayodele pouncing for a touchdown as the Saints took a 17-0 lead.
The victory sends New Orleans to the top of NFC South with a 4-0 record while the Jets slip to 3-1 in AFC East alongside the Patriots.
The defensive-minded Broncos maintained their perfect start to the campaign with a 17-10 home victory in Denver over the Dallas Cowboys.
All the Cowboys points came in the first quarter before the Broncos, who had only conceded 16 points in their first three games, shut down the visitors.
The Broncos took the lead on Brandon Marshall's 51-yard touchdown catch from Kyle Orton with 1:46 remaining.
Orton threw two touchdowns in all as his team went 4-0 - their best start in six seasons - while counterpart Tony Romo was sacked five times by Denver's top-ranked defense.
The Cowboys now stand at 2-2.
A week after losing starting quarterback Chad Pennington for the season, the Miami Dolphins secured their first win of the campaign by defeating the Buffalo Bills 38-10.
Stand-in Chad Henne threw one touchdown but it was the rushing of Ronnie Brown and Ricky Williams that got the job done at Land Shark Stadium.
Brown ran for 115 yards and two touchdowns while Williams added 85 yards and one score.
The injury-plagued Bills gave up 31 points in a 24-minute span with Trent Edwards sacked six times and throwing three interceptions.
The St Louis Rams remain winless after a fourth straight loss, going down 35-0 in San Francisco to the 49ers.
Tight end Vernon Davis caught a 13-yard pass from Shaun Hill for his third touchdown in two weeks, while Patrick Willis returned an interception 23 yards for a score.
Josh Morgan also caught a 24-yard touchdown pass from Hill early in the fourth quarter before Ray McDonald scooped up a fumble by Rams quarterback Kyle Boller and ran 11 yards for another 14 seconds later.
Making his first start for St Louis, Boller went 13 for 24 for 108 yards and was sacked five times in place of injured starter Marc Bulger.
The result was the 49ers' first shutout in 119 games, their last coming when they beat New Orleans 38-0 on January 6, 2002.
In the late game, reigning Super Bowl champions the Pittsburgh Steelers bounced back from successive defeats to score a 38-28 home win over the San Diego Chargers.
Steelers quarterback Ben Roethlisberger threw 333 yards for two touchdowns in total, Pittsburgh having raced into a 21-0 half-time lead.
The second half belonged to the Chargers and their quarterback Philip Rivers, who threw three touchdown passes.
Jacob Hester also scored on a 41-yard fumble return as San Diego stormed back into contention.
The Steelers held on, though, running back Mewelde Moore passing to Heath Miller in the final quarter for his second touchdown of the night and Jeff Reed kicking an insurance field goal as both teams moved to 2-2 for the season as a result.
