The Broncos defence also remained watertight to help snap a four-game losing streak and move Josh McDaniels' side to a half-game off the top of the AFC West Division.
A series of drives, led by Orton, had seen Denver build up a 16-0 lead - with the Broncos landing three field goals through Matt Prater, while Knowshon Moreno slipped through for his third touchdown of the season.
The Giants cut the deficit to 10 with two field goals from Lawrence Tynes in the third quarter.
However Denver settled the result with Orton's 17-yard touchdown pass to Brandon Stokley, while Prater landed a field goal with five minutes left.
Giants quarterback Eli Manning struggled to make an impact on the night as he was sacked twice by Denver defensive end Elvis Dumervil, who extended his NFL-leading total to 14
Manning coughed up three turnovers in total but did pass for 230 yards.
