EDF Energy Cup holders Ospreys will begin their title defence against Harlequins at the Liberty Stadium.
The Welsh region, who beat Leicester to become the first side from their country to lift the trophy in April, have been drawn in Pool C, alongside Quins, Worcester and London Irish.
Meanwhile, Leicester have been drawn in Pool B and begin their campaign with a trip to Bath, followed by matches against Cardiff Blues and Sale.
A total of 12 Guinness Premiership clubs and four Welsh regions will play in next season's competition, which begins on the weekend of October 4th.
Pool A will be contested by Premiership champions Wasps, Gloucester, Newcastle and Newport-Gwent Dragons, with Pool D featuring Saracens, Bristol, Northampton and Llanelli Scarlets.
