The brothers, who won their first title in Paris in 2003, triumphed 6-4 4-6 7-6 (7-4) in a final which ended with Mahut in floods of tears.
The French pair were playing in only their second major together but after the game went into a deciding set tie-break the Americans secured a vital mini-break at four-all before going on to serve out the match.
The top-seeded duo won their 13th title at the Australian Open in January to go level with the record of John Newcombe and Tony Roche. It was also their 88th career title.
At the age of 35, they now retain the possibility of a career first calendar year Grand Slam.
