World No 1 Djokovic teams up with Ana Ivanovic to represent Serbia while seven-time Grand Slam singles winner Williams partners fellow American John Isner.
After 24 years at the Burswood Dome, the event moves to the new Perth Arena with the eight-team tournament starting on Saturday.
And Djokovic is hoping to use the event as a springboard to the new season, when his main ambition is to win the French Open.
The 25-year-old admitted: "My ambitions are always high. I want to win every Grand Slam that I play.
"But Roland Garros is always at the top of my priority list and ambitions. It's the only Slam I haven't won. I played in the final this year, which is a step further than ever before."
Serbia are the top seeds in Group A and if the seeding goes to plan they would face the United States in the final on January 5.
Each match consists of women's and men's singles followed by mixed doubles.
Group A: Australia (Bernard Tomic, Ashleigh Barty); Germany (Tommy Haas, Andrea Petkovic); Italy (Andreas Seppi, Francesca Schiavone); Serbia (Novak Djokovic, Ana Ivanovic).
Group B: France (Jo-Wilfried Tsonga, Mathilde Johansson); South Africa (Kevin Anderson, Chanelle Scheepers); Spain (Fernando Verdasco, Anabel Medina Garrigues); United States (John Isner, Venus Williams).
