The world No 45 was no match for the Slovakian who won 6-4 6-1 to progress to the last 32 of this prestigious event.
Although Hantuchova, who turns 30 in April, is ranked 30 places below the teenage Briton, she dominated the hard court match winning 10 of the last 12 games.
Hantuchova's big serve proved to be a key weapon against the 19-year-old Londoner who is also earning a reputation for possessing a strong serve.
This was only their second ever meeting, following their Wimbledon clash from 2009 when Robson lost in three sets.
Serena Williams moved within two wins of regaining the world number one ranking after reaching the third round on Tuesday.
The Wimbledon, US Open and Olympic champion showed no obvious signs of physical discomfort during a 6-2 6-1 win over Daria Gavrilova, the 18-year-old former world number one junior from Russia who had reached the main draw as a lucky loser.
"I am very happy to be here," said Williams afterwards. "I feel really good. I played a really tough girl who has been junior champion, so I think it was a really good match."
Mona Barthel shocked number five seed Angelique Kerber 6-1 6-2 to reach round three, where she is joined by Maria Sharapova and Marion Bartoli.
Russia's Sharapova dispatched French qualifier Caroline Garcia 6-3 6-2, while world No.11 Bartoli overcame Francesca Schiavone 7-6 (7/5) 6-3.
Other winners in the first round were Nadia Petrova, Roberta Vinci, Lucie Safarova, Sloane Stephens, Ekaterina Makarova, Zakopalova, Hsieh Su-Wei, Sorana Cirstea, Urszula Radwanska, Christina McHale, Monica Niculescu, Romina Oprandi, Daniela Hantuchova and Timea Babos.
