The world No 1 has opted for the relaxed exhibition event at Stoke Park in Buckinghamshire ahead of full ATP tournaments in Eastbourne and Hertogenbosch.
The Serbian first featured at Stoke Park in 2006, and he beat Andy Murray in straight sets in last year's competition.
He said: "I'm so happy to be coming back to The Boodles. It's always great preparation for Wimbledon for me because I know that the quality of the tennis and the opponents is always really high.
"Stoke Park is a beautiful place to play and the crowd always makes it lots of fun for us too."
Djokovic joins a stellar field including Juan Martin Del Potro, Tomas Berdych, Richard Gasquet and defending champion Marin Cilic for the five-day event, which starts on June 18.
