Peng, a semi-finalist in her only previous appearance at the clay-court event in 2010, has been undergoing treatment on a shoulder injury since a second-round defeat in Miami more than a month ago.
However, the 27-year-old shrugged off her lack of preparation to see off the erratic Bartoli, who double-faulted on eight occasions as her serve was broken five times.
"I"m very happy to win in such windy conditions," said world No 34 Peng. "The clay was blowing around a lot, making it even more difficult."
World No 14 Bartoli, who has now lost her last four WTA matches, said: "The first match on clay is always tough for me. I had a lot of trouble finding my game and my feet on the court."
Third-seed Anastasia Pavlyuchenkova of Russia beat Shahar Peer of Isreal 6-4 6-4, but German eighth seed Julia Goerges lost to Monica Puig of Puerto Rico 7-6 (7-3) 6-2.
Wimbledon quarter-finalist Tamira Paszek has not won since the Australian Open and the 31st-ranked Austrian suffered her seventh straight defeat as she went down 6-1 6-3 to Romanian Monica Niculescu.
