The world No 36 was coming off three straight first-round losses on clay but showed that grass is more to her liking at Devonshire Park.
The resounding win, in one hour and 16 minutes, was defined by Vesnina's ability to capitalise once she got into position to break serve.
American qualifier Hampton, who faces a crunch Wimbledon first-round clash against compatriot Sloane Stephens, could not build on her impressive semi-final victory over former world No 1 Caroline Wozniacki, but it was not for the lack of opportunities.
She had five break chances against the Vesnina serve, but the 23-year-old squandered them all, whereas her 26-year-old opponent seized on four of seven openings.
The eventual crushing margin of victory allowed Vesnina to get her hands on a trophy that has been previously won by some of the biggest names in tennis, including Martina Navratilova, Monica Seles, Lindsay Davenport, Justine Henin and Kim Clijsters.
Her first title of the year, gained in Hobart in January, was also the first of Vesnina's 11-year career after losing her first six WTA finals. She will now move up to 22nd in the world as a result of her success.
"I told myself I just needed to stay in the match, try to play every single point," said Vesnina, who who won the French Open doubles title with Ekaterina Makarova last month. "Don't think about wind and just fight. It was just about fighting spirit.
"I'm pretty happy about my game today with these tough conditions."
Vesnina will now begin her Wimbledon campaign against Czech world No 95 Andrea Hlavackova, in their first career meeting.
