World number 51 Granollers, winner in 2011 when he beat fellow Spaniard Fernando Verdasco in the final, upset his Czech opponent 6-2 6-4 in just 63 minutes to progress through to the second round.
Granollers will next face either defending champion Thomaz Bellucci or Federico Delbonis.
Another former winner here was not so lucky though as 2007 champion Paul-Henri Mathieu of France was knocked out by sixth-seeded Russian Mikhail Youzhny 6-3 4-6 6-2.
Fifth seed Feliciano Lopez, runner-up here to Richard Gasquet in 2006, also progressed through, the Spaniard defeating Czech Jan Hernych 7-6 (7/2) 6-3.
In Monday's other match Andrey Kuznetsov beat Sergiy Stakhovsky 7-5 3-6 6-2.
Stakhovsky has now lost four successive matches since his stunning win over Roger Federer at Wimbledon.
