The Frenchman, who can still qualify for the ATP World Tour Finals in London with a strong finish to the season, broke once in each set against 6ft 10in serve-volley specialist Karlovic.
"When you are playing with a hard server like Karlovic you should be very careful when you get a chance to break," said Gasquet.
"Today, I had only two break points and I managed to convert both. Meanwhile, I managed to save a break point on my serve and that made the difference."
Gasquet will play Mikhail Kukushkin in the final after the Kazakh qualifier surprised second seed and defending champion Andreas Seppi in his semi-final.
Kukushkin, the world No.102, came through 6-1 1-6 6-4 in one hour and 38 minutes.
