Second seed Robredo, who received a first-round bye, was looking to win the tournament for a second time in four years but he came unstuck against Argentinian Leonardo Mayer, with the world number 91 claiming a 6-3 6-7 (6/8) 6-4 win in two hours 20 minutes.
Granollers, the fourth seed, was also an early casualty in Vina del Mar as he was defeated 7-6 (9/7) 7-6 (7/2) by Colombian Santiago Giraldo in his first outing of the competition.
It was a better day for another Spanish seed as number seven Guillermo Garcia-Lopez progressed, although he was also given a scare before overcoming Italian Paolo Lorenzi 3-6 7-5 6-4 to secure his spot in the quarter-finals, where he will face Giraldo.
Fifth seed Jeremy Chardy also avoided an upset as the Frenchman defeated home hope Christian Garin (Chi) 7-5 6-0 in his first-round match.
Also in the opening round, Argentinian Horacio Zeballos beat Chilean Gonzalo Lama 6-3 6-3.
