Despite taking the first set, the 32-year-old Finn, who reached the final at last year's event where he was eventually beaten by Juan Monaco, suffered a 6-7 (5/7) 6-3 6-2 loss to Istomin in just under two hours.
World No 41 Monaco of Argentina saw off Benjamin Becker with a comfortable 6-2 6-1 victory to set up a second-round clash with Spanish second seed Marcel Granollers, who received a bye.
Russia's Nikolay Davydenko progressed thanks to a straight-sets victory over Dudi Sela of Israel. The former world No 3 moved past Sela with a simple 6-3 6-1 win in 60 minutes.
Qualifier Mate Delic of Croatia reached the second round thanks to a hard-fought 7-5 7-5 triumph over Poland's Michal Przysiezny.
The one outstanding first-round match, between qualifiers Alessandro Gianessi and Jason Kubler, was suspended at one-set all due to bad light. The pair will resume first on court three on Wednesday, with the winner to face Istomin later in the day.
