Town captain Scott Cuthbert had a looping header cleared off the line, while Luton took the lead on 34 minutes as Dan Potts' cross-shot reached Marriott who beat Cisak from close range.
Marriott should have had a second, shooting wide when unmarked in the area, while Orient threatened a leveller, Ollie Palmer firing straight at Tyler.
After the break, Jonathan Smith fired over, but Orient were level on 47 minutes as Sean Clohessy crossed for Simpson to score.
Cameron McGeehan tried his luck from range, clawed away by Cisak, while Bradley Pritchard could not prod home at the far post and Tyler saved with his legs from Sammy Moore's volley.
Marriott had a great chance to win it in the final 10 minutes, sent clean through by Luke Guttridge, only to drag wide
