Leading scorer Luc Castaignos put Twente ahead in the 55th minute and then, after Vitesse had lost Patrick van Aanholt to a red card on the hour mark, the hosts wrapped up the win through Dusan Tadic's strike 12 minutes from time.
The result sees Twente move to within a point of Ajax, who play sixth-placed Heerenveen on Sunday.
For fourth-placed Vitesse, the defeat means they have now gone five matches without a win.
Elsewhere, AZ Alkmaar remain in seventh spot after only being able to draw 1-1 at home with FC Utrecht, Aron Johannsson's 21st-minute opener for the hosts being cancelled out by Jens Toornstra's leveller 10 minutes into the second half.
At the other end of the table, ADO Den Haag climbed off the bottom as a 53rd-minute Mitchell Schet strike saw them claim a 1-0 win at Roda JC, who now prop up the standings.
NEC Nijmegen are also out of the bottom two after beating RKC Waalwijk 3-1 away from home.
RKC took a sixth-minute lead through Romeo Castelen but NEC were ahead by the break thanks to goals from Navarone Foor (25) and Lasse Nielsen (39) before Michael Higdon (72) wrapped up the win in the second half.
