Siem De Jong opened the scoring for Ajax after just four minutes before a second-half brace from Lerin Duarte - either side of Ricardo Kishna's goal - added gloss to the scoreline.
With Twente and Feyenoord starting the day second and third in the table respectively, Ajax knew they would distance at least one of their rivals but it ended up being both as Twente came from 2-0 down to claim a draw at De Grolsch Veste.
Feyenoord had gone ahead through Graziano Pelle in the 14th minute with Jean-Paul Boetius adding a second 10 minutes into the second half.
But after Luc Castaignos got one back in the 68th minute, substitute Cuc Martina snatched a point at the death.
The draw allowed Vitesse Arnhem to move into third place, one point above Feyenoord, with a 3-1 win over struggling RKC Waalwijk.
The visitors took a shock lead through Kenny Anderson in the 14th minute and it took until the second half for Vitesse to restore order.
But Mike Havenaar's 50th-minute equaliser was swiftly followed by Valeri Qazaishvili's goal and Christian Atsu made sure in the 68th minute.
Substitute Johan Martensson scored the only goal in the 88th minute as FC Utrecht edged out FC Groningen 1-0.
