The American pair birdied the final six holes in the better-ball format to move to 19-under with a round to go at Tiburon Golf Club in Florida.
First-round leaders Davis Love III and Brandt Snedeker fired a seven-under 65 but that dropped them from two in front to two behind.
Jason Dufner and Vijay Singh, Stewart Cink and Carl Pettersson, and Charles Howell III and Rory Sabbatini are all tied for third at 15-under.
Defending champions Keegan Bradley and Brendan Steele shot a 64 to get to 13 under.
The leaders' six-birdie burst started with a chip-in from O'Hair on 13 before a huge downpour and strong winds delayed play for one hour and 50 minutes.
But, unpeturbed, Perry birdied 14 and 15 when play resumed, O'Hair birdied 16 and 17, and Perry added yet another on the last hole.
"We got pretty hot," O'Hair said. "Kenny did a great job when I wasn't in the hole ... then I did a pretty good job when Kenny wasn't in the hole."
The 12 two-man teams will play a scramble on Sunday. The winners will split $750,000.
