O'Loughlin, who last played at Wembley in August in the Challenge Cup Final, has been out with an Achilles injury, but has been given the all-clear to play after seeing a specialist on Monday.
Warriors head coach Shaun Wane told the Wigan website: "Sean has had a good week and it is great news, he is fit to play."
Wolves head coach Tony Smith is looking forward to a clash of styles when his side meets their local rivals in the showpiece event for the first time since 1990.
Smith has named an unchanged squad of 19 from last week's semi-final win over the Huddersfield Giants.
"Both are well-supported clubs so it should be a great atmosphere and an intriguing game too as there's a contrast in styles; we are probably a bit more adlib and Wigan are a bit more structured in their attacking style," Smith told the Wolves website.
"The fact that we've had such competitive games over the last two or three years also makes it intriguing."
