Jamie Murray and John Peers lost 6-4 6-4 to the French pairing of Nicolas Mahut and Pierre-Hugues Herbert in Saturday's US Open Men's Doubles final.
It is the second time this year that Murray and Peers have finished as runners up in a Grand Slam following the pair's Wimbledon final defeat earlier this summer.
And they will be left to rue missed chances, including an opportunity to break at 4-4 in the first set. Mahut and Herbert responded immediately to that scare to break Peers' serve to love and take the first set.
The second set proved to be as equally tight as the first with neither side able to break the others serve in the first nine games.
However, at 5-4 the French pair stepped up a level and broke Murray for the first time in the match to take the title in straight sets.
"Me and John had a great year to get to the Wimbledon final and then to get to the final here again," said Murray after the match.
"A lot of hard work has gone into this team in the last two-and-a-half years and we'll keep trying to get over the line and get a Grand Slam title."
Australian Peers struggled to hide his disappointment following the defeat.
"It's been a ride, a lot of ups and downs but you've got to look for the positives," he said.
"This is one of the best courts to play on in the world so its been a lot of fun but unfortunately we couldn't win today."
"I am speechless now," said Mahut, who's only previous Grand Slam doubles final ended in defeat at the French Open in 2013.
"We have been working so hard for so many years to win this kind of tournament. We lost in the final and I had another loss two years ago. We said today, 'let's go for it' and we played great.
"Pierre played great in the last game. I have never seen anyone play like that in a doubles match - unbelievable shots."
Herbert added: "He deserves it. He came to me last year and said, 'come on, let's play together, I believe in you'.
"I am so happy to win a grand slam and that he can win a Grand Slam, so thank you Nico for everything."
