In the annual match between PGA Tour players who own houses or reside on either one of the two Florida golf estates, England's Ian Poulter and Justin Rose beat Australian Nick O'Hern and American JB Holmes in the final match of the day to edge Lake Nona ahead after the rival clubs had matched each other 4-4 in the first four matches of the two-day tournament at Isleworth.
World number one Woods revealed he had considered competing in the event, but later claimed he had not had sufficient time to prepare for competition following his recent four-month hiatus from the sport.
Instead he will now make his comeback at next month's Masters at Augusta National.
On Monday, teams of two played each other in four-ball, betterball matches over 18-holes with the winning pair of each match picking up two points.
However, on Tuesday a head-to-head singles format will be used with a singles win being worth two points and with 20 points at stake in the ten matches.
Aussies Stuart Appleby and Robert Allenby of Isleworth, with a five-under-par score, narrowly defeated South Africans Ernie Els and Trevor Immelman in the first match of the day to take their team into the lead.
Isleworth pulled further ahead when the American duo Mark O'Meara and John Cook beat England's Ross Fisher and American Ben Curtis.
But Lake Nona hit back when Northern Ireland's Graeme McDowell and Sweden's Peter Hanson went seven-under to beat India's Arjun Atwal and US veteran Lee Janzen.
The visitors then squared the match as South Africa's Retief Goosen and Sweden's Henrik Stenson produced a sparkling eight-under display to ease past England's Brian Davis and American Charles Howell III.
That set up an intriguing finish to the day and ultimately Rose and Poulter went round in seven-under-par to clinch their round-grabbing win.
Isleworth leads the six-year-old rivalry 3-2 with one tournament drawn.
