Dallas quarterback Tony Romo suffered a broken collarbone but replacement Brandon Weeden was able to steer the Cowboys to a 20-10 victory over the Philadelphia Eagles.
Romo, 35, was injured in the third quarter on a play in which he was sacked and fumbled. He suffered a similar injury in week seven in 2010 and missed the rest of the season.
The Cowboys, rated as one of the favourites to reach the NFL play-offs, are already without star receiver Dez Bryant, who broke a small bone in his foot in the season-opener last week against the New York Giants.
Despite the injury blow, Kyle Wilber returned a blocked punt 26 yards for a touchdown while Weeden threw a 42-yard touchdown pass to Terrance Williams as the Cowboys overcame a team-record 18 penalties to record their 10th consecutive road win.
Philadephia running back DeMarco Murray had two yards rushing on 13 carries in his first game against his former team.
The Cowboys moved their record for the season to 2-0 while the Eagles slipped to 0-2.
In the other late kick-offs on Sunday afternoon, Oakland Raiders quarterback Derek Carr threw a 12-yard touchdown pass to Seth Roberts with 26 seconds left against the Baltimore Ravens to cap the most productive day of his young career in a 37-33 win.
Carr threw for a career-high 351 yards and three touchdowns.
And Jacksonville Jaguars' kicker Jason Myers slotted a 28-yard field goal with 40 seconds left in regulation to give his side a nail-biting 23-20 win over the Miami Dolphins.
