Former Huddersfield and Hull stand-off Chris Thorman led the spree with a record 54 points from four tries and 19 goals, while former Keighley half-back Jonny Presley scored six of his side's 23 tries.
Thorman's personal tally topped the previous individual best by Hull KR's George West, who scored 53 points against Brookland Rovers in 1905, while York's score beat the previous record of 119 set by Huddersfield against Swinton Park in 1914.
Huddersfield still hold the record for the biggest win in any game thanks to their 142-4 victory over Blackpool Gladiators in a Regal Trophy tie in 1994 while Barrow beat Nottingham City 138-0 in the same competition in 1994.
Comfortable
On a day of high-scoring matches, Widnes centre Steve Tyrer and Batley scrum half Gareth Moore both scored 26 points to help their clubs to comfortable third-round victories.
